Damages in pelvic floor muscles often cause dysfunction of the entire urogenital system, which is clinically challenging. A bioengineered skeletal muscle construct that mimics structural and functional characteristics native could provide a therapeutic option to restore normal function. However, most current constructs are unable timely innervation necessary for successful grafting recovery. We previously have demonstrated post-synaptic acetylcholine receptors (AChR) clusters can be pre-formed on cultured myofibers with agrin treatment suggested implantation AChR containing accelerate recovery In this study, we develop 3-dimensional (3D) bioprinted human construct, consisting multi-layers bundles aligned myofibers, investigate effect efficiency vivo. Agrin successfully vitro increased neuromuscular junction (NMJ) formation vivo transposed nerve model rats. rat injury, resulted reconstruction accelerated innervation. This approach may solution many challenges associated resulting from lack suitable tissue efficient function restoration.
